Both of them are part of what are called "adjetivos posesivos" (possessive adjectives, not sure if that's how you call them in English too), and since they are adjectives, they modify nouns and they have to match in number with the noun they are modifying. "Tu" is singular and "tus" is plural. So, you'd say "tu perro" to mean "your dog" and "tus perros" to mean "your dogs". Imagine as if you were saying "yours dogs" in the second case, that's how we use "tus".
